Pune Highway is a crime-thriller set in a Mumbai suburb building known as 'Shantibhavan' where Khandu, Vishnu, Nicky, Natasha, and Babu all grow up together. Life in 'Shantibhavan' is still wonderful despite Babu becoming paraplegic due to an incident on the Pune Highway. Life seems to be all fun until a body is discovered floating in a lake 200 kilometers distant one day. Identifying the body takes time. However, when they do, it causes the friendships to fall apart in several ways.
