Holy Cow is a satirical comedy set in a small town where Salim Ansari (Sanjay Mishra) stays with his wife Safiya Ansari (Sadiya Siddiqui) and has a cow named Rukhsar that goes missing one day. On knowing that his cow has gone missing his wife Sadiya asks him to search and get it back home. Salim along with his friend Rambo (Mukesh Bhatt) try to find it without letting people know that he is a Muslim and didn’t mean to get the cow home to kill and have it as meat because killing cows is not treated sacred among Hindus. What does Salim do to get back his cow forms the remaining part of this interesting tale.
